Kumamoto Tokyu REI Hotel is the one we would book for a multi-night stay in the city, and the reason is dull and decisive: there is a guest coin laundry. Rates start around ¥7,900 for two, and it sits by the Karashimacho tram stop, about ten minutes from JR Kumamoto Station by tram and inside walking distance of the arcades and Kumamoto Castle. The gap to the cheapest of the three central chains is about ¥900 a night — that is what the laundry costs you, and on a single night it earns nothing, so book Daiwa Roynet Hotel Kumamoto instead if you are moving on in the morning.
What a coin laundry is worth on a Kyushu trip
Kyushu trips tend to be loops. Fukuoka, Kumamoto, Kagoshima, maybe Takachiho or Aso, over a week or ten days, with a Shinkansen or a rental car threading them together. The hotel is at Karashimacho, which is one of the two useful stops in central Kumamoto. From JR Kumamoto Station the tram takes about ten minutes. That is the number to hold onto, because Kumamoto separates its Shinkansen station from its city centre, and a lot of visitors book the wrong one.
Karashimacho puts you next to Sakuramachi Bus Terminal, where the airport limousine arrives and where long-distance buses leave. It also puts the Shimotori and Kamitori arcades and Kumamoto Castle within a walk. For a stay where you go out at night and come back on foot, this is the right part of town.

Who this hotel is not for
It is a business hotel, and it behaves like one. Rooms are compact, the design is functional, and nothing about the stay is an experience in itself. If you want a hotel that is part of the trip rather than a place to sleep in the middle of it, this is the wrong tier — Kumamoto has full-service options for about half as much again.
It is also the wrong choice for a single night before an early Shinkansen. A ten-minute tram ride is nothing in the afternoon and an irritation at 06:30 with luggage. For that trip, book on the station side and accept a duller evening.
How it fits a Kumamoto itinerary
The case for this hotel is strongest on a stay of three nights or more, and stronger still if Aso is on your list.
